It doesn't look like he is asking how to remove the battery. He is asking about charging the device.
To answer your question, it doesn't matter either way. Whether you charge with a usb power adapter or with your computer, the device can be off or on.

To piggy back onto Matt's answer, it doesn't matter.
If you've ever charged a normal iPod (one with a screen) via the usb port on a computer, your iPod will sync with the computer, so it doesn't matter if it was on or off.
